MAIN CONCLUSION: This study compared the plastomes of Ulmaceae allowing analyses of the dynamic evolution, including genome structure, codon usage bias, repeat sequences, molecular mutation rates, and phylogenetic inferences. Ulmaceae is a small family in the order Rosales. This family consists of seven genera, including Ulmus, Zelkova, Planera, Hemiptelea, Phyllostylon, Ampelocera, and Holoptelea. Ulmaceae is an interesting lineage from plant biogeographic, systematic, evolutionary, and paleobotanic perspectives. It is also a good model to investigate the evolution of the plastomes in woody plants. In this study, we sequenced and assembled the complete plastomes of the six Ulmaceae genera to compare genomic structures and reveal the molecular evolutionary patterns. The size of the quadripartite plastomes ranged from 158,290 bp to 161,886 bp. The genomes contained 131 genes, including 87 coding genes, 36 tRNA, and 8 rRNA. The gene number, gene content, and genomic structure were highly consistent among the Ulmaceae genera. Nine variable regions including ndhA intron, ndhF-rpl32, ycf1, psbK-trnS, rps16-trnQ, trnT-trnL, trnT-psbD, trnS-trnG, and rpl32-trnL, were identified in Ulmaceae plastomes according to the nucleotide diversity values. Condon usage was biased among the genes and showed consistent trends in the seven genera. Molecular evolution analyses revealed that most of the genes and all gene groups were under widespread purifying selection. Twelve genes (ccsA, matK, psbH, psbK, rbcL, rpl22, rpl32, rpoA, rps12, rps15, rps16, and ycf2) were under positive selection. Phylogenetic analyses supported that Ulmaceae should be divided into two main clades, such as the temperate clade, including Ulmus, Zelkova, Planera, and Hemiptelea and the tropical clade, including Phyllostylon, Ampelocera and Holoptelea. This study reports the structure and evolutionary characteristics of the Elm family. These new genomic data will benefit assessments of genomic evolution and provide information to elucidate the phylogenetic relationships among Ulmaceae species.

Ulmus lanceifolia, which plays an important role in ecology and economy, is one of the main evergreen broad leaved arbor species in southern China. In this study, we assembled its complete chloroplast genome. The total genome size of U. lanceifolia was 158,652bp, containing a large single-copy region of 87,119bp, a small single-copy region of 18,697bp, and a pair of inverted repeat regions of 26,418bp. The overall GC content of U. lanceifolia chloroplast genome is 35.63%. We obtained the 132 genes chloroplast genome, including 86 proteincoding genes, 8 rRNA genes, and 37 tRNA genes. The phylogenetic analysis suggests that U. lanceifolia is a monophyletic taxa and a transitional species between the 2 groups which were combined by Ulmus americana, U. elongate and Zelkova serrate, Z. schneideriana respectively in Ulmaceae.

Zelkova schneideriana Hand.-Mazz. of the Ulmaceae family is a Tertiary relict and economically deciduous tree species endemic to Central and Southern China. In this study, we performed a transcriptome sequencing of Z. schneideriana using high-throughput sequencing approach to detect polymorphic expressed sequence tag-simple sequence repeats (EST-SSR) markers. A total of 3,235 microsatellite loci were detected from 53,517 unigenes. A set of 30 microsatellite markers were randomly selected to validate in 41 individuals from three populations, of which 10 were polymorphic. The number of alleles per locus ranged from 3 to 11. The observed heterozygosity and expected heterozygosity ranged from 0.366 to 0.829 and 0.439 to 0.848, respectively. These polymorphic SSR primers showed good transferability across different Zelkova species, and are valuable for future studies on genetic diversity, conservation, phylogeography, and species delimitation in Z. schneideriana, as well as other Zelkova species. PREMISE: The hydraulic architecture in the leaves, stems and roots of plants constrains water transport and carbon gain through stomatal limitation to CO2 absorption. Because roots are the main bottleneck in water transport for a range of plant species, we assessed the ecophysiological mechanism and importance of a high fraction of root hydraulic resistance in woody and herbaceous species. METHODS: Biomass partitioning and hydraulic conductance of leaves, stems, and roots of Japanese knotweed (Fallopia japonica, a perennial herb) and Japanese zelkova (Zelkova serrata, a deciduous tall tree) were measured. Theoretical analyses were used to examine whether the measured hydraulic architecture and biomass partitioning maximized the plant photosynthetic rate (the product of leaf area and photosynthetic rate per leaf area). RESULTS: Root hydraulic resistance accounted for 83% and 68% of the total plant resistance for Japanese knotweed and Japanese zelkova, respectively. Comparisons of hydraulic and biomass partitioning revealed that high root-resistance fractions were attributable to low biomass partitioning to root organs rather than high mass-specific root conductance. The measured partitioning of hydraulic resistance closely corresponded to the predicted optimal partitioning, maximizing the plant photosynthetic rate for the two species. The high fraction of root resistance was predicted to be optimal with variations in air humidity and soil water potential. CONCLUSIONS: These results suggest that the hydraulic architecture of plants growing in mesic and fertile habitats not only results in high root resistance due to small biomass partitioning to root organs, but contributes to efficient carbon gain.

A novel compound 1 and nine known compounds (2-10) were isolated by open column chromatography analysis of the root bark of Ulmus davidiana. Pure compounds (1-10) were tested in vitro to determine the inhibitory activity of the catalytic reaction of soluble epoxide hydrolase (sEH). Compounds 1, 2, 4, 6-8, and 10 had IC50 values ranging from 11.4 ± 2.3 to 36.9 ± 2.6 µM. We used molecular docking to simulate inhibitor binding of each compound and estimated the binding pose of the catalytic site of sEH. From this analysis, the compound 2 was revealed to be a potential inhibitor of sEH in vitro and in silico. Additionally, molecular dynamics (MD) study was performed to find detailed interaction signals of inhibitor 2 with enzyme. Finally, compound 2 is promising candidates for the development of a new sEH inhibitor from natural plants.

Bioactivity-driven LC/MS-based phytochemical analysis of the root bark extract of Ulmus davidiana var. japonica led to the isolation of 10 compounds including a new coumarin glycoside derivative, ulmusakidian (1). The structure of the new compound was elucidated using extensive spectroscopic analyses via 1D and 2D NMR spectroscopic data interpretations, HR-ESIMS, and chemical transformation. The isolated compounds 1-10 were tested for their antifungal activity against human fungal pathogens Cryptococcus neoformans and Candida albicans. Compounds 9 and 10 showed antifungal activity against C. neoformans, with the lowest minimal inhibitory concentration (MIC) of 12.5-25.0 µg/mL, whereas none of the compounds showed antifungal activity against C. albicans.

RESUMO

A preliminary phytochemical investigation of the stems of the endangered plant Ulmus elongata led to the isolation of a new coumarin derivative (named ulmuselactone A, 1) and eight known compounds (2-9). The new structure was elucidated by detailed analysis of comprehensive spectroscopic methods, and its absolute configuration was established by comparing experimental and calculated electronic circular dichroism (ECD) spectra. The isolated compounds were evaluated for their antibacterial activities.

Ulmus elongata is a species of Sect. Chaetoptelea (Liemb.) S chneid in Ulmaceae, and it is an endangered wild plant listed in the second class of the Protected Plants in China. The complete chloroplast genome (cp) of U. elongata was reported in this study. The result showed that the cp genome was 159,230 bp in length including a large single-copy (LSC) 87,718 bp and a small single-copy (SSC) 18,690 bp, which were separated by two inverted repeats (IRs) of 26,411 bp with the typical quadripartite structure, respectively. The genome encoded 132 genes, including 87 protein-coding genes, 37 tRNA genes, and eight rRNA genes. The GC content was 35.57%. Chloroplast sequences were used for constructing phylogenetic tree to determine the evolutionary status of U. elongata. The maximum-likelihood phylogenetic analysis showed that U. elongata was clustered with five other Ulmus species, and the relationship between Ulmus and Zelkova was closest. The success of cp genome assembly of U. elongata has laid a foundation for the study of chloroplast molecular biology and can effectively promote the study of genetic breeding and molecular evolution of U. elongata.

The complete chloroplast genome sequence of the Tertiary relict tree Zelkova serrata was reported in this study. The chloroplast genome is 158,875 bp in length with a typical angiosperm quantitative structure consisting of a large single copy (87,412 bp) and a small single copy (18,683 bp) separated by a pair of inverted repeat (26,390 bp). Genome annotation revealed a total of 129 genes comprising 84 protein-coding genes, 37 tRNA genes, and eight rRNA genes. Phylogenomic analysis based on the whole plastomes indicated that Z. serrata and Z. schneideriana formed a well-supported monophyletic clade sister to genus Ulmus in Ulmaceae.

PREMISE: Seed dispersal is extremely important for the recovery and restoration of forest communities. Relict tree genus Zelkova possesses a unique dispersal mechanism: mature fruits fall with the entire twig, and the dried leaves that are still attached function as a drag-enhancing appendage, carrying the fruits away from the parent tree. This singular adaptation has never been investigated in Z. abelicea. METHODS: Drop tests with dispersal units and individual fruits of Z. abelicea were performed in controlled conditions to measure their dispersal velocity and to define their flight mode. RESULTS: Zelkova abelicea uses both slowly falling dispersal units with chaotic motion, as well as fast falling individual fruits using a straight path. The falling velocity of Z. abelicea dispersal units is 1.53 m s-1 , which is virtually identical to that of the East Asiatic Z. serrata (1.51 m s-1 ). In contrast, the falling velocity of individual fruits was 2.74 m s-1 (Z. serrata: 5.36 m s-1 ). CONCLUSIONS: Members of the genus Zelkova, growing today in distant regions, show remarkable evolutionary conservation of the velocity and flight mechanics of their dispersal units. This is surprising because the Mediterranean and East Asiatic Zelkova species have been separated at least 15-20 mya. Zelkova abelicea, although growing in the Mediterranean with completely different forest structure and composition, still uses the same dispersal mechanism. The dispersal capacity of the genus Zelkova is less efficient than that of other wind dispersed trees, and it presumably evolved for short-distance ecological spread and not for long-distance biogeographical dispersal.

PREMISE: Pteroceltis tatarinowii (Ulmaceae), the only species of the genus Pteroceltis, is an endangered tree in China. Here, novel expressed sequence tag-simple sequence repeat (EST-SSR) markers were developed to illuminate its genetic diversity for conservation and assisted breeding. METHODS AND RESULTS: Based on Illumina transcriptome data from P. tatarinowii, a total of 70 EST-SSR markers were initially designed and tested. Forty-eight of 70 loci (68.6%) were successfully amplified, of which 20 were polymorphic. The number of alleles per locus ranged from two to six, and the levels of observed and expected heterozygosity ranged from 0.018 to 0.781 and from 0.023 to 0.702, respectively. Additionally, cross-amplification was successful for 17 loci in two related species, Ulmus gaussenii and U. chenmoui. CONCLUSIONS: These new EST-SSR markers are valuable transcriptomic resources for P. tatarinowii and will facilitate population genetics and molecular breeding of this species and its relatives in Ulmaceae.

Ulmus parvifolia Jacq is a kind of landscape tree endemic to East Asia. In this study, the complete chloroplast genome of U. parvifolia was sequenced. The genome was 159,259 bp in length, with a large single-copy (LSC) region of 88,451 bp, a small single-copy (SSC) region of 19,598 bp, and two inverted repeat (IR) regions of 25,605 bp, each. The genome consisted of 121 genes, including 77 protein-coding genes, 36 tRNA genes, and 8 rRNA genes. The phylogenetic results indicated that Ulmus is not monophyletic and U. parvifolia constitute a well-supported clade sister to U. Sect. Ulmus. The complete chloroplast genome of U. parvifolia will provide important information for phylogenetic and evolutionary studies in Ulmaceae, as well as the other closely related families.

The root bark of Ulmus davidiana var. japonica (Ulmaceae), commonly known as yugeunpi, has been used as a traditional Korean medicine for the treatment of gastroenteric and inflammatory disorders. As part of continuing projects to discover bioactive natural products from traditional medicinal plants with pharmacological potential, phytochemical investigation of the root bark of this plant was carried out. This led to the successful isolation of a new chromane derivative (1) and 22 known compounds: catechin derivatives (2-5), megastigmane glycoside (6), dihydrochalcone glycosides (7 and 8), flavanone glycosides (9 and 10), coumarins (11 and 12), lignan derivatives (13-17), and phenolic compounds (18-23). The structure of the new compound (1) was determined with 1D and 2D NMR spectroscopy and HR-ESIMS, and its absolute configurations were achieved by chemical reactions and the gauge-including atomic orbital (GIAO) NMR chemical shifts calculations. All the isolated compounds were evaluated for their potential biological activities including neuro-protective, anti-neuroinflammatory, and anti-Helicobacter pylori activities. Among the isolates, compounds 1, 8, and 20 displayed stronger potency by causing a greater increase in the production and the activity of nerve growth factor (NGF) in C6 glioma cells (147.04 ±â€¯4.87, 206.27 ±â€¯6.70, and 143.70 ±â€¯0.88%, respectively), whereas compounds 11, 14, and 19 inhibited nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated murine microglial cells (IC50 of 18.72, 12.31, and, 21.40 µM, respectively). In addition, compounds 1, 11, 18, and 20 showed anti-H. pylori activity with MIC values of 25 or 50 µM against two strains of H. pylori 51 and 43504. These findings provide scientific evidence that supports the traditional usage of U. davidiana var. japonica root bark in the treatment of gastroenteric and inflammatory disorders.

We provide the first report of Tischeriidae on Ulmaceae, a novel host-plant family, and describe Tischeria ulmella Xu Dai, sp. nov., a new species discovered in the Yellow River Scenic Area of Zhengzhou (Henan Province, China), feeding on Ulmus pumila L. We also provide a redescription of the Quercus-feeding Tischeria naraensis Sato discovered in China for the first time. Both species are illustrated with photographs of the adults, male and female genitalia, and the leaf mines.

BACKGROUND: Literature survey proved the use of the powdered sun-dried bark and roots of Celtis africana for the treatment of cancer in South Africa. OBJECTIVE: The aim of this study was to do further isolation work on the ethyl acetate fraction and to investigate the cytotoxic activities of the various fractions and isolated compound. MATERIALS AND METHODS: Cytotoxicity of petroleum ether, chloroform, ethyl acetate, n-butanol fractions and compound 1 were tested on mouse lymphoma cell line L5178Y using the microculture tetrazolium assay. RESULTS: One new glucosphingolipid 1 was isolated from the aerial parts of C. africana. The structure of the new compound was determined by extensive analysis by one-dimensional and two-dimensional nuclear magnetic resonance spectroscopy and mass spectrometry. The ethyl acetate fraction and compound 1 showed strong cytotoxic activity with an EC50 value of 8.3 µg/mL and 7.8 µg/mL, respectively, compared with Kahalalide F positive control (6.3 µg/mL). CONCLUSION: This is the first report of the occurrence of a cytotoxic glucosphingolipid in family Ulmaceae.

UNLABELLED: • PREMISE OF THE STUDY: The Caucasus represents one of the world's biodiversity hotspots and includes the climatic refugia Hyrcan on the southern coast of the Caspian Sea and Colchis on the eastern coast of the Black Sea, where different species survived during the Quaternary climatic oscillations. We evaluated the genetic diversity of the relict tree Zelkova carpinifolia shared between the two refugia and distributed throughout the Caucasus and adjacent areas.• METHODS: Specimens were collected from 30 geographical sites in Azerbaijan, Georgia, Iran, and Turkey and screened for variability at eight nuclear microsatellite loci. The genetic diversity among and within populations was assessed using a set of statistical measures.• KEY RESULTS: We detected 379 different genotypes from a total of 495 individuals with varying degrees of clonal reproduction at the different sites. Low to intermediate levels of genetic diversity were observed at all sites, and strong differentiation between sampling sites was absent. In addition, we observed no clear genetic differentiation between the Colchis and Hyrcan. Bayesian clustering of the genotypes revealed three populations with high levels of admixture between the sampling sites.• CONCLUSIONS: The lack of strong genetic structure of studied populations of Z. carpinifolia contrasts with a previous study based on chloroplast markers and suggests that long-distance pollen dispersal is an important factor of gene flow among populations of Z. carpinifolia. The present study does not reveal any particular site with particularly isolated genotypes that would deserve more attention for conservation purposes than others, although some sites should be considered for further investigation.

PREMISE OF THE STUDY: The current study aimed at developing nuclear microsatellite markers for the relict tree species Zelkova carpinifolia, which is threatened in its natural range in the South Caucasus. • METHODS AND RESULTS: Pyrosequencing of an enriched microsatellite library on the Roche FLX platform using the 454 Titanium kit produced 86,058 sequence reads, most of which contained short tandem repeats. Eighty microsatellite loci identified using the software package QDD version 1 were selected and tested for proper PCR amplification. Of these, 13 allowed proper amplification and were shown to be polymorphic among a sample of 25 Z. carpinifolia specimens from various geographic origins. • CONCLUSIONS: The set of microsatellite markers will be suitable for the assessment of genetic diversity in Z. carpinifolia. They will allow for an examination of phylogeographic patterns as well as of population structure and gene flow within this species.

Medicinal plants are used by traditional practitioners to treat several ailments. Ethnomedicinal studies on Trema orientalis Linn. Blume (Ulmaceae) have shown that it is used in the treatment of diabetes mellitus, respiratory diseases, oliguria, and malaria. This article is aimed at providing comprehensive information on the medicinal uses, biology, phytochemical constituents, and pharmacological data available on T. orientalis. This has been done to explore its therapeutic potential for future research opportunities. This review was compiled with information obtained from databases such as Medline, Elsevier, Springer, Science Direct, Pubmed, Google Scholar, and a library search for articles published in peer-reviewed journals. Compounds present in the plant include tannins, saponins, flavanoids, triterpenes, phytosterols, and several constituents of xanthones. Some pharmacological research done on the plant has focused on, hypoglycemic activity, analgesic, anti-inflammatory activities, anti-plasmodial activity, diuretic activity, laxativity effect, anti-convulsant activity, anti-helmintic activity, anti-sickling effect, anti-oxidant, and anti-bacterial activity. This compilation strongly supports the view that T. orientalis has beneficial therapeutic properties, and indicates its potential as an effective herbal remedy for several diseases. The promising results from several research works could be further substantiated by clinical trials.

A espécie Celtis iguanaea (Jacq.) Sargent é popularmente conhecida como esporão de galo ou grão de galo. As folhas são indicadas pelo uso popular para o tratamento de dores no corpo e no peito, para reumatismo, asma, cólicas, má digestão e como diurético; as raízes são utilizadas para infecções urinárias e as cascas para a febre. O presente trabalho objetivou contribuir para o estudo fitoquímico e atividade fitotóxica com enfoque alelopático das cascas de Celtis iguanaea. O extrato etanólico foi submetido à partição com os solventes hexano, clorofórmio e acetato de etila. As substâncias friedelina e epifriedelinol (triterpenos) foram isoladas da fração hexano e identificadas por meio de métodos espectroscópicos de RMN de ¹H e 13C. O extrato bruto na concentração de 0,1 mg mL-1 causou inibição acentuada do hipocótilo em 34,97% e estimulou o crescimento da radícula em 29,64% de plântulas de Lactuca sativa. No ensaio de toxicidade frente à Artemia salina o extrato bruto e frações apresentaram uma CL50 superior a 1000 μg mL-1, indicando que o mesmo não possui efeito tóxico.


The species Celtis iguanaea (Jacq.) Sargent is popularly known as "esporão de galo" or "grão de galo". Its leaves are recommended by the popular use for the treatment of body and chest aches, as well as for rheumatism, asthma, cramps, indigestion and as diuretic; its roots are used for urinary infections and its bark for fever. This study aimed to contribute to the phytochemical investigation of the toxic activity focused on the allelopathic effect of the bark of Celtis iguanaea. The ethanol extract was subjected to solvent partition with hexane, chloroform and ethyl acetate. The substances friedelin and epifriedelinol (triterpenes) were isolated from the hexane fraction and identified by spectroscopic methods ¹H and 13C NMR. The crude extract at a concentration of 0.1 mg mL-1 caused marked inhibition of hypocotyl in 34.97% and stimulated radicle growth in 29.64% seedlings of Lactuca sativa. In the toxicity test against Artemia salina the crude extract and fractions showed an LC50 higher than 1000 μg mL-1, indicating that it has no toxic effect.

Vinte e cinco caprinos, criados em piquetes, morreram nos cinco dias subsequentes ao fornecimento de folhas de Trema micrantha (fam.Ulmaceae), uma árvore com nome comum de grandiúva. Quatro caprinos foram necropsiados e amostras de vísceras foram coletadas para exame histológico. As principais alterações clínicas foram: apatia, anorexia, cabeça apoiada contra obstáculos, decúbito e morte. Achados macroscópicos incluíram sufusões no epicárdio e endocárdio; fígado levemente amarelado e com padrão lobular evidente e, em um caso, acompanhado de hemorragias multifocais. Na histologia observou-se necrose hepática, que variava de centrolobular a massiva, compatível com hepatopatia tóxica. No SNC havia satelitose, tumefação neuronal, edema periaxonal, perivascular. O diagnóstico de intoxicação por Trema micrantha foi baseado no quadro clínico e lesional de hepatite tóxica associado ao uso da planta para alimentação de caprinos.


Twenty five goats, maintained in paddocks, had died in five subsequent days after have been offered leaves, mixed in the ration, of Trema micrantha, a tree commonly called grandiúva. Four animals were necropsied and samples were collected for histopathology. Clinical signs included apathy, anorexia, head pressing against obstacles, decubitus and death. Macroscopic findings included suffusions in the epi- and endocardium, a yellowish liver with pronounced lobular pattern, in one goat, the liver presented additionally multiple visible hemorrhages. Histological examination revealed centrilobular to massive hepatic necrosis consistent with acute liver toxicosis. In the brain, satelitosis, neuronal swelling, and perineuronal and perivascular edema was found. The diagnosis of poisoning by Trema micrantha was based in the clinical picture characteristic of toxic hepatitis associated in the feeding of the plant to the goats.
